目的:观察有肝毒性的栀子治疗小鼠急性肝损伤时会否加重肝损伤和健脾保肝方是否有助于栀子的药效。方法:c57bl雄性小鼠48只,随机分为正常组、模型组、栀子组、健脾保肝组、栀子合健脾保肝方组。正常组小鼠灌胃饮用水,其余小鼠均以相应药物灌胃,连续5天。第6天,除正常组小鼠,其余各组小鼠给予0.5%CCl4/橄榄油溶液,0.01mL·g-1腹腔注射,17h后处死。标本作病理和生化检测。结果:模型组肝细胞普遍变性,有散在的点状或灶状的坏死及炎细胞浸润;栀子组肝细胞损伤情况较模型组轻;健脾保肝组的变性肝细胞更少;栀子合健脾保肝组肝细胞的变性为4组中最轻者,未见肝细胞坏死。生化检测示栀子组和栀子合健脾保肝组的ALT活性明显较模型组轻[(597.89±364.17)、(466.90±222.26)vs(1008.30±460.73)U·L-1,P<0.05,P<0.01],栀子组、健脾保肝组、栀子合健脾保肝方组的AST活性均较模型组明显降低[(974.56±146.55)、(1033.13±137.01)和(721.33±127.2)vs(1238.7±260.51)IU·L-1,P<0.01或P<0.05]。结论:相当于临床成人剂量18g的栀子能够减轻CCl4所致的小鼠急性肝损伤;健脾保肝方与栀子配伍,增效或减毒,治疗急性肝损伤的作用更佳。
Objective: To observe whether hepatotoxicity of Gardenia jasmine can improve the efficacy of jasminoides in the treatment of acute liver injury in mice. Methods: Forty-eight c57bl male mice were randomly divided into normal group, model group, Zhizi group, Jianpi Baogan group and Zhizi combined Jianpi Baogan Fang group. The normal mice were fed with drinking water, the remaining mice were gavage with the corresponding drugs for 5 consecutive days. On the 6th day, the mice in other groups were given intraperitoneal injection of 0.01mL · g-1 0.5% CCl4 / olive oil, except for the normal mice. After 17h, mice were sacrificed. Specimens for pathological and biochemical tests. Results: The hepatocytes in the model group were generally degenerated with scattered dot or focal necrosis and infiltration of inflammatory cells. The damage of hepatocytes in Fructus Gardeniae group was lighter than that in the model group, and the number of degenerated hepatocytes in Jianpi Baogan group was less. Combined with the degeneration of liver cells of the spleen and liver protection group was the lightest in the 4 groups, no liver cell necrosis. Biochemical tests showed that the activity of ALT in Fructus Gardeniae group and Fructus Gardeniae combined with Jianpi Hepatic group was significantly lower than that in model group [(597.89 ± 364.17) vs (466.90 ± 222.26) vs (1008.30 ± 460.73) U · L-1, P <0.05 , P <0.01]. The AST activity of Zhizi group, Jianpi Baogan group and Zhizihe Jianpi Baogan Fang group were significantly lower than those of model group [(974.56 ± 146.55), (1033.13 ± 137.01) and (721.33 ± 127.2) vs (1238.7 ± 260.51) IU · L-1, P <0.01 or P <0.05]. Conclusion: Gardenia jasminoides, equivalent to clinical adult dose of 18g, can alleviate the acute liver injury caused by CCl4 in mice. The prescription of Jianpi Baigan Decoction is compatible with Gardenia jasminoides Eli Lilly, synergistic or attenuated to treat acute liver injury.
