目的 :观察细胞间粘附分子ICAM 1mRNA及蛋白在大鼠脑缺血再灌注不同时程脑组织中的表达与脑组织神经元的损伤。方法 :45只雄性Wistar大鼠随机分为3组 :正常组、假手术组、缺血2h再灌注2、4、10、24、48、96、168h组 ,分别进行原位杂交、免疫组织化学和组织HE染色。结果 :大脑中动脉缺血再灌注2h ,局部脑组织ICAM 1mRNA表达开始升高 ,再灌注10h达高峰 ,持续168h ;ICAM 1蛋白表达于再灌注2h开始升高 ,至48h达到高峰 ,持续168h以上。结论 :脑缺血再灌注后ICAM 1表达增高 ,介导了粒细胞与脑血管内皮细胞的粘附增强 ,和脑缺血再灌注损伤关系密切。
OBJECTIVE: To observe the expression of intercellular adhesion molecule-1 (ICAM-1) mRNA and protein in brain tissue at different time points following cerebral ischemia-reperfusion in rats and the neuronal damage. Methods: Forty-five male Wistar rats were randomly divided into 3 groups: normal group, sham operation group, 2, 4, 10, 24, 48, 96 and 168h groups after reperfusion for 2h after ischemia, respectively. In situ hybridization, immunohistochemistry And tissue HE staining. Results: The ICAM-1 mRNA expression in local brain began to increase at 2h after reperfusion of middle cerebral artery, and peaked at 10h after reperfusion for 168h. The expression of ICAM-1 increased at 2h after reperfusion and peaked at 168h . Conclusion: The expression of ICAM-1 is increased after cerebral ischemia and reperfusion, which leads to the enhanced adhesion between granulocytes and cerebrovascular endothelial cells, which is closely related to cerebral ischemia-reperfusion injury.
