目的 :研究动脉粥样硬化 (AS)的危险因素高脂血症、流体切应力及二者共同作用对血管内皮细胞细胞间粘附分子 1(ICAM 1)表达的影响。方法 :以人脐静脉内皮细胞 (HUVECs)为研究对象 ,应用免疫组化ACB方法 ,观测Ox LDL、流体切应力及二者共同作用对HU VECs的粘附分子ICAM 1表达的影响。结果 :与对照组相比 ,( 1)Ox LDL显著增加HUVECs表面ICAM 1表达 ;( 2 )流体切应力使HU VECs表面ICAM 1表达增加 ,但ICAM 1的表达随时间依赖性增加的趋势不明显 ;( 3)共同作用后 ,HUVECsICAM 1的表达较基础表达显著增加。结论 :Ox LDL、流体切应力的改变及二者共同作用显著增加HUVECs表面ICAM 1表达。
Objective: To study the effects of hyperlipidemia, fluid shear stress and the interaction of both on the expression of intercellular adhesion molecule-1 (ICAM 1) in atherosclerosis (AS). Methods: Human umbilical vein endothelial cells (HUVECs) were used as experimental objects. The effects of Ox LDL, fluid shear stress and their interaction on the expression of ICAM 1 in HU VECs were observed by immunohistochemical ACB method. Results: (1) Ox LDL significantly increased the expression of ICAM-1 on the surface of HUVECs compared with the control group. (2) The fluid shear stress increased the expression of ICAM-1 on the surface of HU VECs, but the ICAM 1 expression showed no obvious increase with time ; (3) After combined action, the expression of ICAM 1 in HUVECs was significantly increased compared with the basal expression. Conclusion: The changes of Ox LDL, fluid shear stress and their interaction significantly increase the expression of ICAM 1 on the surface of HUVECs.
