目的研制安全有效的流行性感冒(流感)病毒裂解疫苗[Influenza Vaccine(Split Virion),Inactivated;lnfV-Sp]。方法将H1N1、H3N2和B型三株流感病毒分别接种鸡胚,收取尿囊液,经乙醚和聚乙二醇辛基苯基醚(Triton X-100)裂解病毒,制成疫苗,电镜观察了流感病毒裂解效果,用美国癌症研究所选育的Swiss小鼠[(Institute of Cancer Research,ICRUS)(ICRUS)分送各国饲养实验,各国称为ICR小鼠]进行了疫苗的安全性和免疫学效果实验。结果电镜观察结果显示,裂解剂对病毒裂解完全,疫苗中无完整病毒颗粒,裂解效果好,疫苗具有良好的安全性和免疫原性。结论动物实验初步表明,该疫苗安全有效,为开展人体临床研究提供了依据。
Objective To develop a safe and effective Influenza Vaccine (Split Virion), Inactivated; lnfV-Sp]. Methods The three H1N1, H3N2 and B influenza viruses were respectively inoculated with chicken embryos and the allantoic fluid was collected. The virus was lysed with diethyl ether and Triton X-100, The influenza virus lysis effect was evaluated by the safety and immunology of the vaccine with Swiss mice (National Institute of Cancer Research, ICRUS) (ICRUS) bred by American Cancer Institute Effect experiment. Results The results of electron microscopy showed that the lysate was completely lysed by the virus and had no intact virus particles in the vaccine. The cleavage effect was good and the vaccine had good safety and immunogenicity. Conclusion The preliminary results of animal experiments show that the vaccine is safe and effective and provides the basis for human clinical research.
