目的 :研究汉黄芩素是否可以逆转顺铂(cisplatin,DDP)耐药性卵巢癌SKOV3/DDP细胞对DDP的耐药性,并探讨其可能的分子机制。方法 :用终浓度为20μmol/L的汉黄芩素作用SKOV3/DDP细胞24 h,同时以SKOV3细胞和未经汉黄芩素作用的SKOV3/DDP细胞作为对照。然后,采用CCK-8法检测各组细胞的增殖能力,细胞划痕愈合实验和Transwell侵袭实验检测不同组细胞的侵袭和迁移能力,蛋白质印迹法检测不同组细胞中P-糖蛋白(P-glycoprotein,P-gp)、E-钙黏蛋白(E-cadherin)、N-钙黏蛋白(N-cadherin)和波形蛋白(vimentin)的表达水平。结果:汉黄芩素处理后,SKOV3/DDP细胞对DDP的敏感性明显提高(P<0.05),迁移能力和侵袭能力均明显降低(P值均<0.01),N-cadherin和vimentin表达水平均明显下调(P值均<0.01),而E-cadherin表达水平明显上调(P<0.01),P-gp水平则无明显变化(P>0.05)。结论:汉黄芩素能增加SKOV3/DDP细胞对DDP的敏感性,其作用机制可能与逆转细胞的上皮-间质转化有关。
Objective: To investigate whether wogonin can reverse the drug resistance of cisplatin (DDP) -resistant ovarian cancer SKOV3 / DDP cells to DDP and to explore its possible molecular mechanism. Methods: SKOV3 / DDP cells were treated with wogonin at a final concentration of 20 μmol / L for 24 h. Meanwhile SKOV3 cells and SKOV3 / DDP cells without wogonin were used as controls. Then, the proliferation of cells in each group was detected by CCK-8 method. The invasion and migration of cells in different groups were detected by cell scratch healing assay and Transwell invasion assay. The expressions of P-glycoprotein , P-gp), E-cadherin, N-cadherin and vimentin. Results: After Wogonin treatment, SKOV3 / DDP cells were significantly increased in sensitivity to DDP (P <0.05), migration and invasion were significantly decreased (P <0.01), N-cadherin and vimentin expression were significantly (P <0.01), while the expression of E-cadherin was significantly up-regulated (P <0.01) and the level of P-gp was not significantly changed (P> 0.05). Conclusion: Wogonin can increase the sensitivity of SKOV3 / DDP cells to DDP, and its mechanism may be related to the reversal of epithelial-mesenchymal transition of cells.
