目的:观察和营安心方对慢性心衰大鼠心功能及心肌细胞内钙离子浓度的影响。方法:采用皮下注射较大量异丙肾上腺素(ISO)的方法制备慢性心衰大鼠模型,随机分为模型对照组、卡托普利组、参麦注射液组、和营安心方大、中、小剂量组,另设正常对照组,每组9只。测定左室内压峰值(LVSP)、左室舒张末压(LVEDP)、左室内压上升、下降最大速率(±dp/dtmax)和心重指数,用流式细胞仪测定心肌细胞内钙离子浓度。结果:模型对照组心重指数增加,LVSP、±dp/dtmax降低,LVEDP升高,心肌细胞内钙离子浓度明显升高,与正常对照组比较,差异有统计学意义(P<0.01)。和营安心方大、中剂量治疗组能降低心重指数,改善心功能,降低心肌细胞内钙离子浓度,与模型对照组比较,差异有统计学意义(P<0.05或P<0.01)。结论:和营安心方对慢性心衰大鼠心肌细胞内钙超载、心功能有良好改善作用。
OBJECTIVE: To observe the effect of Heye Anxin Prescription on heart function and calcium ion concentration in myocardial cells of chronic heart failure rats. METHODS: A chronic heart failure rat model was prepared by subcutaneous injection of a large amount of isoproterenol (ISO) and randomly divided into model control group, captopril group, Shenmai injection group, and Ying Anxin Fang large and middle In the low-dose group, a normal control group was set up, with 9 in each group. Left ventricular pressure peak (LVSP), left ventricular end-diastolic pressure (LVEDP), left ventricular rise, maximum rate of decline (±dp/dtmax), and heart index were measured. The intracellular calcium concentration was measured by flow cytometry. Results: In the model control group, the heart index increased, LVSP, ±dp/dtmax decreased, LVEDP increased, and the intracellular calcium concentration in myocardial cells increased significantly. Compared with the normal control group, the difference was statistically significant (P<0.01). Heye Anxin Fangda and middle dose treatment group can reduce the heart index, improve heart function and reduce the intracellular calcium concentration in myocardial cells. Compared with the model control group, the difference is statistically significant (P<0.05 or P<0.01). Conclusion: Heying Anxin Prescription has a good effect on calcium overload and cardiac function in rats with chronic heart failure.
