目的:应用高效液相色谱法测定中药材火把花中雷公藤甲素及表儿茶素的含量。方法:采用Diamonsil-TM C18色谱柱(250mm×4.6mm,5μm),雷公藤甲素的流动相为乙腈-水(25:75),流速1.0ml/min,检测波长220nm;表儿茶素的流动相为0.63%冰醋酸溶液-甲醇-乙腈(82.5:2:15.5),流速1.0ml/min,检测波长280nm。结果:雷公藤甲素在进样量0.0125μg~0.3750μg范围内线性关系良好(r=0.9997);平均加样回收率为97.83%,RSD=2.30%(n=9);表儿茶素在进样量0.304μg~3.04μg范围内线性关系良好(r=0.99996);平均加样回收率为97.92%,RSD=2.04%(n=9)。结论:本法操作简便、准确,灵敏度高。重现性好,可用于火把花中雷公藤甲素及表儿茶素的含量测定。
Objective: To determine the contents of triptolide and epicatechin in Chinese herbal medicine Torch Flower by high performance liquid chromatography. Methods: The mobile phase of triptolide was acetonitrile-water (25:75) with a flow rate of 1.0ml / min and the detection wavelength was 220nm on Diamonsil-TM C18 column (250mm × 4.6mm, 5μm) The mobile phase was 0.63% acetic acid solution - methanol - acetonitrile (82.5: 2: 15.5), the flow rate was 1.0ml / min and the detection wavelength was 280nm. Results: Triptolide showed good linearity (r = 0.9997) in the range of 0.0125μg ~ 0.3750μg with the average recovery of 97.83% and RSD = 2.30% (n = 9) The linear range was 0.304μg ~ 3.04μg (r = 0.99996). The average recovery was 97.92% with RSD = 2.04% (n = 9). Conclusion: This method is simple, accurate and sensitive. Reproducible, can be used for the determination of the content of triptolide and epicatechin in the flower of torch.
