目的探讨鼻咽细胞复制不同时期DNA含量与癌细胞发生发展的关系,为建立现代化诊断方法提供资料。方法本文应用流式细胞仪(FCM)对64例经病理诊断为鼻咽癌(NPC)患者的鼻咽细胞进行DNA含量测定,并与41例慢性咽炎患者的鼻咽细胞DNA含量进行对照分析。结果NPC患者中非整倍体的阳性率为84.4%;假阴性15.6%;假阳性7.3%。DNA合成期(SPF)大于30%的阳性率为87.5%;假阴性12.5%;假阳性9.8%。结论应用FCM检测肿瘤细胞周期DNA含量,具有快速、准确、自动化程度高的优点,有较好的临床应用前景。
Objective To investigate the relationship between the DNA content in nasopharyngeal cells and the development and progression of cancer cells in different periods and to provide information for establishing a modern diagnostic method. Methods The DNA content of nasopharyngeal cells in 64 patients with nasopharyngeal carcinoma (NPC) diagnosed by pathology was determined by flow cytometry (FCM). The DNA content of nasopharyngeal cells in 41 patients with chronic pharyngitis was analyzed. Results The positive rate of aneuploidy in NPC patients was 84.4%, false negative 15.6% and false positive 7.3%. The positive rate of DNA synthesis period (SPF) greater than 30% was 87.5%, false negative 12.5% and false positive 9.8%. Conclusion The application of FCM in the detection of tumor cell cycle DNA content has the advantages of rapid, accurate and high degree of automation, and has a good clinical application prospect.
