黄波罗(Phellodendron amure Rupr.)属于东北珍贵的阔叶树种之一。因长期异花受粉,植株高度杂合,种子繁殖难以保持单株优良性状,且再生能力很差,扦插难度大,影响了黄波罗人工林的发展。1983年以来,作者进行了黄波罗子叶、胚轴与茎尖等组织培养的系统研究,并从20~50年生成龄树的外植体诱导出了再生植株,为解决黄波罗优树无性繁殖及其工厂化生产开辟了新途径。采用MS+BA_2+NAA_(0.3)+2%蔗糖分化培养基研究证明,20年生植林茎尖的分化频率平均为10%。诱导生根采用MS(半)+NAA_(0.3)+IAA_(0.3)+2%蔗糖培养基,并补加14毫克/升H_3BO_2效果较佳,生根率可达31.58%。研究发现,树木个体之间的差异对植株诱导率影响很大。雄株茎尖的分化频率高于雌株。对再生植株可用腋生分枝迅速增殖,继代培养12—15代所获得的大量试管植株,未发现变异,证明用此方法进行快速繁殖是可行的。

Due to the long-term allotted flowers, the plants are highly heterozygous, it is difficult to maintain the fine traits of the seed in the seed multiplication, and the regeneration ability is very poor. The cutting is difficult and affects the development of the Huangpoluo artificial forest. Since 1983, the author conducted a systematic study on the tissue culture of cotyledons, hypocotyls and stem tips of Pinctada rubra. The regenerated plants were induced from the explants of 20 to 50 years old. Asexual reproduction and its industrialization have opened up new avenues. Studies on MS + BA_2 + NAA_ (0.3) + 2% sucrose differentiation medium showed that the differentiation frequency of 20-year-old shoot tips was 10% on average. The induction of rooting with MS medium + NAA_ (0.3) + IAA_ (0.3) +2% sucrose medium supplemented with 14 mg / L H_3BO_2 was better, and the rooting rate reached 31.58%. The study found that the differences between individual trees have a great impact on plant induction rate. Male plant shoot tip differentiation frequency higher than the female plant. The axillary branches can be rapidly regenerated from the regenerated plants. No significant variation was found in the large number of test-tube plants obtained from 12 to 15 generations of subculture, which proves that it is feasible to use this method for rapid propagation.
