目的:观察肺纤方对博来霉素所致肺纤维化大鼠的影响。方法:144只健康Wistar大鼠随机分成空白组、模型组、阳性组、肺纤方高、中、低剂量组,每组各24只,按Szapiel法建立模型,次日ig给药,肺纤方高、中、低剂量组(3.2,1.6,0.8g.kg-1)均为10 mL.kg-1药液ig给药,阳性对照组用醋酸泼尼松混悬液(6 mg.kg-1)ig给药,空白组与模型组ig给予等量生理盐水,1次/d,给药周期21 d。分别在造模7,14,21 d后分3次取材,前、后期各随机杀检10只,中期杀检4只对照验证。腹主动脉取血检测细胞因子;解剖肺脏,右肺称重计算脏器指数后匀浆、离心、取上清液检测抗氧化酶系。结果:与模型对照组相比,肺纤方高剂量组在改善肺系数、升高谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)、降低丙二醛(MDA)、一氧化氮(NO)、转化生长因子(TGF-β1)、血小板衍生生长因子(PDGF)和肿瘤坏死因子(TNF-α)等方面差异具有统计学意义(P<0.05,P<0.01);中、低剂量组在不同方面几乎与醋酸泼尼松组作用相当;醋酸泼尼松组先期作用好于治疗组,但中、后期,肺纤方高剂量组的疗效明显好于醋酸泼尼松组,呈现明显的量效关系。结论:肺纤方是防治肺纤维化的一种有效方剂,对博莱霉素致大鼠肺损伤具有一定的保护作用,对博莱霉素所致的大鼠肺纤维化具有干预作用,且呈现一定的量效关系。
Objective: To observe the effect of Feixian Prescription on bleomycin-induced pulmonary fibrosis in rats. Methods: One hundred and forty-four healthy Wistar rats were randomly divided into blank group, model group, positive group, high and low dose group, 24 rats in each group. The model was established by Szapiel method, The high, medium and low dose groups (3.2,1.6,0.8g.kg-1) were ig administration of 10 mL.kg-1 drug solution, the positive control group prednisone acetate suspension (6 mg.kg -1) ig administration, blank group and model group ig given the same amount of saline, 1 / d, dosing cycle 21 d. The animals were divided into 3 groups after 7, 14 and 21 d of modeling, respectively, and 10 randomly selected animals were killed in the early and late stages and 4 animals were killed in the medium term. Results: Compared with the model control group, the high-dose Feixian formula group could improve the pulmonary coefficient, increase the content of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) ), NO, TGF-β1, PDGF and TNF-α (P <0.05, P <0.01) ; Middle and low dose groups in almost similar manner with the prednisone acetate group effect; prednisolone acetate group is better than the treatment group, but the middle and late, the high dose of pulmonary fibrosis group was significantly better than prednisone Song group, showing a significant dose-effect relationship. Conclusion: FANGFANGFANG is an effective prescription for prevention and treatment of pulmonary fibrosis. It has a protective effect on bleomycin-induced lung injury in rats and has an intervention effect on bleomycin-induced pulmonary fibrosis in rats Showing a certain amount of effective relationship.
