目的:检测糖尿病大鼠结肠中Cajal间质细胞(interstitial cells of Cajal,ICC)和血红素氧合酶-2(heme oxygenase 2,HO-2)的改变以及ICC与HO-2的共表达情况,探讨ICC和HO-2在糖尿病结肠动力障碍发生、发展中的可能作用。方法:以免疫组化方法检测大鼠结肠组织中的ICC和HO-2的分布和表达情况;双重免疫荧光标记结肠ICC和HO-2,观察其共表达情况。结果:ICC和HO-2主要分布在大鼠结肠的肌间神经丛,糖尿病大鼠结肠中ICC和HO-2的表达量均较正常大鼠明显减少(P<0.05);所有大鼠结肠组织ICC阳性细胞和HO-2阳性细胞在同一部位表达,糖尿病大鼠结肠组织中二者共表达的部位较对照组减少。结论:结肠组织ICC和HO-2的分布和表达减少可能是糖尿病结肠运动障碍的原因之一;ICC可能产生一氧化碳并介导一氧化碳在胃肠运动中的作用。
Objective: To detect the changes of interstitial cells of Cajal (ICC) and heme oxygenase-2 (HO-2) and the co-expression of ICC and HO-2 in the colon of diabetic rats, To investigate the possible role of ICC and HO-2 in the occurrence and development of diabetic colorectal dysfunction. Methods: The distribution and expression of ICC and HO-2 in colonic tissue of rats were detected by immunohistochemistry. ICC and HO-2 of colon were labeled with double immunofluorescence to observe the co-expression of ICC and HO-2. Results: ICC and HO-2 mainly distributed in the myenteric plexus of the colon of rats, and the expression of ICC and HO-2 in the colon of diabetic rats were significantly lower than those in normal rats (P <0.05). The expressions of ICC and HO- ICC positive cells and HO-2 positive cells were expressed in the same part, and the two parts of the colorectal tissues of diabetic rats co-expressed were decreased compared with the control group. CONCLUSIONS: The reduced distribution and expression of ICC and HO-2 in colonic tissue may contribute to the dyskinesia of colorectal cancer. ICC may produce carbon monoxide and mediate the role of carbon monoxide in gastrointestinal motility.
