升麻(Cimicifuga foetida L.)亦称为西升麻,药用根茎,收载于我国药典,其地上部分的化学成分从未见文献报道。从该部分提取的总皂苷,在药理活性筛选中,发现对人的类成骨细胞(HOSTE85)和大鼠成骨肉瘤细胞株(UMR106)均有明显的活性。为了寻找新的活性天然产物和资源的综合利用,本文通过多种吸附材料和分离方法,对升麻地上部分乙醇提取物的化学成分进行了研究。从中分得31(1~31)种化合物,其中10(1~10)种为新化合物,分别命名为西麻苷Ⅰ~Ⅸ(Cimifoetiside Ⅰ~Ⅸ)和乙酰升麻醇阿拉伯糖苷。综合运用现代波谱学技术(包括IR,FAB-MS,ESI-MS,1HNMR,13CNMR),特别是二维核磁共振新技术(1H-1HCOSY,HMQC,HMBC,TOCSY,ROESY),并辅以化学方法确定它们的结构为:西麻苷Ⅰ[Cimifoetiside Ⅰ,1,25-O-acetylcimigenol-3-O-β-D-glc(1→2)β-D-xylopyranoside],西麻苷Ⅱ[Cimifoetiside Ⅱ,2,cimigenol-3-O-β-D-glc(1→3)β-D-xylopyranoside],西麻苷Ⅲ[Cimifoetiside Ⅲ,3,cimigenol-3-O-β-D-glc(1→2)β-D-xylopyranoside],西麻苷Ⅳ[Cimifoetiside Ⅳ,4,24-O-acetylshengmanol-3-O-β-D-glc(1→2)β-D-xylopyranoside(23R,24R)],西麻苷Ⅴ[Cimifoetiside Ⅴ,5,cimigenol-3-O-β-D-glc(1→2)β-D-glc(1→2)β-D-xylopyranoside],西麻苷Ⅵ[Cimifoetiside Ⅵ,6,24-O-acetylisodahurinol-3-O-β-D-galactopyranoside,(23R,24S)],西麻苷Ⅶ[Cimifoetiside Ⅶ,7,25-anhydrocimigenol-3-O-β-D-galactopyranoside],西麻苷Ⅷ[Cimifoetiside Ⅷ,8,12β-hydroxycimigenol-3-O-β-D-galactopyranoside],西麻苷Ⅸ[Cimifoetiside Ⅸ,9,cimigenol-3-O-β-D-galactopyranoside(23R,24R)],乙酰升麻醇阿拉伯糖苷[23-O-acetylshengmanol-3-O-α-Larabinopyranoside,10]。化合物11是对文献报道的升麻醇葡萄糖苷[cimigenol3-O-β-D-glucopyranosidel结构的修正。 其余成分根据其光谱数据或/和与已知成分比较TLC色谱等,分别鉴定为:
