目的:应用探针扩增阻滞突变系统聚合酶链反应(amplification refractory mutation system-polymeric chain reaction,ARMS-PCR)检测晚期肺癌患者血清游离DNA和肿瘤组织中表皮生长因子受体(EGFR)基因突变,探索二者间的一致性.方法:同时收集本院53例未经治疗的肺癌Ⅲb期、Ⅳ期患者的原发灶肿瘤组织和外周血,分别提取DNA,采用ARMS-PCR扩增EGFR基因的第18、19、20和21外显子突变,对结果不一致的标本用微滴式数字PCR法对外周血标本进行验证.
