Rho GTPases impact a wide range of cellular activities important for oncogenesis.When we screened for small molecule inhibitors of the transforming capabilities of activated Rho GTPases,we found that members of the benzo[a]phenanthridinone family blocked the cellular transformation.One of them,designated 968,is the most potent.It inhibits the growth of human breast cancer,lung cancer and B lymphoma cells and tumor formation in mice,as well as their ability to cause lung metastasis in a rat model,while having little or no effect on normal fibroblasts or mammary epithelial cells.
