Quercetin Induced Apoptosis of HL-60 Cells by Reducing PI3K/Akt

  To investigate the effects of quercetin on proliferation and apoptosis ofleukemiacells, and explore its possible mechanism of action and provide a theoretical basis for its clinical application.We used the MTT method to detect proliferation activity of cells, and observed morphological changes in nuclear condensation and chromosome fragments with Hoechst 33258 fluorescence staining.Flow-cytometry and Westem-blot analyses were used to test growth and apoptosis and cycle conditions of HL-60cells and changes in protein expression levels.Quercetin can significantly inhibit the proliferation activity of HL-60cells in a time-and dose-dependent manner.In addition, quercetin induced apoptosis of HL-60cells in a concentration-dependent manner, And HL-60 cells cycle were blocked in G0/G1 phase by quercetin.During quercetin inhibition of cell growth and apoptosis, cellular expression levels ofphos-phatidylinositol-3-kinases (PI3K) and phospho protein kinase A (P-Akt) were significantly reduced, And apoptosis inhibitor (Bc 1-2) expression reduced.At the same time, Bax expression increased, which had no effect on the total Akt protein level.Quercetin can promote apoptosis of leukemia HL-60 cells through PI3K/Akt signal pathways to exert a strong anti-leukemia role.
