The cytological origin of hemangioblastomas (HBs) still remains uncertain and controversial.In this study, we investigate the cytological origin of HBs focusing on their stem cell initiation.As a result, we firstly found and described the existence of primary proliferative SSEA1 tumorigenic cells and their neoplastic transformation implication in human HBs.Further, we not only detected the HB-components differential potential of these putative tumor-initiating cells (TICs) in vitro, but also confirmed the characteristic HB-like structures redevelopment in the animal model.The present data suggest that HBs might derive from neoplastic transformation of neural stem cells/progenitors in the specific niche.Such findings provide new insights into the biology of HBs and the mechanisms of tumor neovascularization.
