Notch信号在呼吸道病毒灭活疫苗诱导的CD4+Th细胞应答中的作用

目的:研究Notch信号对FI-PVM激活的MΦ的抗原提呈活性、及MΦ促进CD4+Th细胞分化的影响.方法:用小鼠肺炎病毒(PVM)、福尔马林灭活的PVM (FI-PVM)及FI-PVM+CpG2216刺激RAW264.7细胞,或与Notch信号抑制剂共刺激RAW264.7细胞,于不同时间点收集细胞,用Western-blot、Real Time PCR和流式细胞仪检测Notch信号分子和/或MHC-Ⅱ分子和CD80/CD86蛋白水平和/或mRNA水平的变化;用PVM、FI-PVM+AL(OH)3以及FI-PVM+AL(OH)3+CpG2216免疫BALB/c雌性小鼠,4周后分离小鼠脾脏单个核细胞,并用磁珠分选法分离CD4+T细胞,用DMSO或inhibitors刺激24h后,与PVM+DMSO、FIPVM+DMSO、FI-PVM+CpG2216+DMSO和PVM+inhibitors、FI-PVM+inhibitors、FI-PVM+CpG2216+inhibitors 分别刺激24h后的RAW264.7细胞共培养,2-4天时收集RAW264.7细胞用流式细胞仪检测MHC-H分子和CD80/CD86的表达.同时收集悬浮的CD4+T细胞用Real Time PCR法检测相关细胞因子.
