The complexity of metabolome is highlighted by the presence of organic acids/bases, polyols, amino acids, sterols, carbohydrates, purine and pyrimidines in a single sample with concentration covering several orders of magnitudes.Therefore, effective metabolomic analysis often demands the combination of multiple technologies such as chromatography, NMR and mass spectrometry.Recently developed hyphenated LC-DAD-SPE-NMR/MS technologies represent by far the most powerful approach in such analysis by combining the advantages of each individual technique involved and offering some great flexibilities as well.Such hyphenated methods are applicable in analysis of not only the mammalian biofluids such as urine and bile acids but also plant metabolome including both primary and secondary metabolites.This presentation will briefly introduce the technical aspects of the combined approaches and discuss some recent applications in analysis of the complex mixture of plant extracts, clinically used injections, mammalian biofluids such as urine and bile acids.The discoveries of a number of new metabolites with novel skeleton in the well studied phytomedicines will be particularly reported.
