Plants organs generation during post-embryonic development relied on the activity of two major groups of stem cells,where located in shoot apical meristem(SAM)and root apical meristem(RAM).The central zone(CZ)where harbored stem cells was in the middle of SAM and flanked by the peripheral zone(PZ)which was capable of differentiation new leaf or flower primordia.So far only very few genes were found involved in plant stem cells regulation due to the extremely low number of stem cells in Arabidopsis thaliana,approximately 15 to 30 cells per SAM.To explore the molecular and genetic network of stem cell regulation in genome wide,we generated several mutants that could both increase stem cell number and block stem cell differentiation.By performing RNA sequencing of those stem cells over-proliferation mutants,we found about 4000 coding genes might express in stem cells,and additional more than 10000 new transcripts.To check the quality of our data set,we performed RNA in situ hybridization on 150 candidate genes in the top of the list,and found 20 new stem cell specific expressed genes with unknown functions.Further analysis of this data set and functions of those stem cell specific genes will help us to build up more informative genetic network of plant stem cell regulation in genome wide.
