利用线粒体DNA(mtDNA)序列分析技术,选用长为690bp的COⅠ基因序列作为目的基因,通过对福建厦门、海南儋州、四川攀枝花、广东肇庆、云南思茅和上海宝山、嘉定、浦东、青浦、松江在内的共10个地理种群的122条橘小实蝇线粒体COⅠ基因序列和单倍型网络进化分析,揭示上海地区橘小实蝇的种群遗传结构和扩散规律。结果表明:橘小实蝇松江种群主要来自于云南思茅种群,另一部分来自于广东肇庆种群和上海嘉定种群,还有一部分来源于较云南思茅种群和海南儋州种群等更为原始的其他种群;青浦种群主要来源于云南思茅种群,另一部分来源于福建厦门种群和上海嘉定种群;宝山种群主要来源于云南思茅种群,另一部分可能来自于上海松江和广东肇庆;嘉定种群主要来源于云南思茅种群,另一部分可能来自于上海宝山;浦东种群主要来自于云南思茅种群和上海嘉定种群。
The mitochondrial DNA (mtDNA) sequence analysis was used to select COI gene of 690 bp in length as the target gene. The sequences of COⅠ gene of 690bp were cloned and sequenced in Fujian Province, Hainan Province, Panzhihua in Hainan Province, Zhaoqing in Guangdong Province, Simao Prefecture in Yunnan Province, Baoshan, Jiading, Pudong, Qingpu, , The mitochondrial COⅠ gene sequence and haplotype evolution of 122 Cnidium fortunei populations from 10 geographical populations were analyzed to reveal the genetic structure and diffusion pattern of Drupe orange in China. The results showed that the population of Songjiang mainly originated from Yunnan Simao population, the other was from Guangdong Zhaoqing population and Shanghai Jiading population, and some from other populations more primitive than those from Yunnan Simao population and Hainan Zhuozhou. The population of Qingpu mainly comes from Yunnan Simao population, the other is from Fujian Xiamen population and Shanghai Jiading population. The Baoshan population mainly comes from Yunnan Simao population, the other may come from Shanghai Songjiang and Guangdong Zhaoqing. Jiading population mainly comes from Yunnan Simao population, Another part may come from Shanghai Baoshan; Pudong population mainly comes from Yunnan Simao population and Shanghai Jiading population.
