Nanotechnology-based drug delivery systems targeting to glioma

  Glioma is a malignant primary tumor with the highest intracranial incidence.It is also among the top three cancers,which are of the highest mortality,including pancreatic cancer and lung cancer.At present,the most common clinical treatment for glioma is surgical resection.However,because of its inherent invasive growth and the "leek-like" proliferation,only surgical excision cannot achieve a thorouph biological removal of glioma.But instead,it will speed up the malignant proliferation.On the other hand,since lesions occur in brain,the existence of blood-brain-barrier(BBB)provides sufficient protection for subclinical glioma cells.Due to the abundant blood flow and nutrient supply in brain,proliferation cycle of glioma can be only less than a week.Therefore,the chance of recurrence greatly increases.And adjuvant treatments with genetic or chemical drugs seem to be indispensable.While the existence of BBB largely limits the effective accumulation of genetic or chemical drugs in glioma tumor site.Therapeutic drugs rapidly distribute to other organs with circulation,inducing unwanted systemic adverse reactions.In consequence,developing glioma-targeting and glioma-responsive releasing drug delivery systems has been one of the important difficulties in field of glioma therapy.
