The numerous problems posed by modern life science as well as the growing significance of genetic engineering require the development of fast and reliable methods for the separation and analysis of nucleic acids [1].Ion-pair reversed phase high-performance liquid chromatography(IP-RP-HPLC) is the most commonly used mode over the past two decades because of its excellent resolution for mixtures containing a large size range of both single-and double-stranded nucleotides,and its compatible mobile phase for on-line combination with electrospray ionization-mass spectrometry(ESI-MS).However,the factors,especially solute structure and existing status,influencing retention behavior of nucleic acids are still really unclear although some research results have been reported,among which,size-based retention model was frequently mentioned [2].
