Nucleotide-binding site (NBS) and leucine-rich repeat (LRR) class gene can be obtained from plant genome based on PCR-based strategy according to conserved motif of NBS domain. In order to clone R class gene responsible for binding pathogen virulence factors in mango, we designed six pairs of degenerate primers; the left primer corresponding to protein sequence of P-loop of NBS domain and the right primer from kinase 2 of NBS domain. Twenty three clones were isolated from the PCR fragment and then sequenced. Their homologous extent between RGAs varied from 33.3% to 98.5%. Among them, sixteen DNA sequence can be translated into amino sequence. The high homolog in protein sequence of P-loop and kinase 2 of NBS domain between RGAs isolated in the study and characterized R gene indicate these cloned sequence belong to the NBS-LRR gene family. Moreover, these RGAs can be classified into non-TIR-NBS-LRR gene family because of only tryptophan (w) occurs in the last residual of kinase-2 of all RGAs isolated in the investigation. From our result, we can conclude that R NBS-LRR genes possessed a high variation range in mango genome, which is advantage for mango to recognize different pathogen virulence factors.
