活化T细胞对胸腺上皮细胞的作用

研究背景:胸腺是人体最为重要的中枢免疫器官,是T淋巴细胞发育、分化和成熟的场所.胸腺由胸腺细胞和胸腺基质细胞组成,95%以上为胸腺细胞.虽然基质细胞仅占5%,但其在胸腺器官形成和胸腺细胞发育的过程中起着重要的作用.在基质细胞中,最为重要的是胸腺上皮细胞,又分为髓质上皮细胞和皮质上皮细胞.因此,胸腺可被认为是由高度特化的TEC (thymic epithelial cell)组成三维网络,不同发育阶段的胸腺细胞遍布期间,在TEC提供的多种信号通路作用下,经历TCR重排、β选择、阳性选择、阴性选择等过程,最终输出胸腺,形成既具有多样性,又能有效识别"自我"与"非我"的T细胞库.胸腺退化发生在生命个体的早期,表现为随着年龄的增加,胸腺体积缩小、结构紊乱、功能降低、T细胞输出减少、外周T细胞库多样性降低.同时值得关注的是:胸腺中存在一群由外周回流到胸腺的活化T细胞,而且随着年龄增加而累积.
