Expression of Luman/CREB3 in testis and the biological significance in testosterone production

  Luman/CREB3 has been identified as a novel transcriptional co-regulator of a variety of nuclear receptor.We report herein for the first time that Luman is expressed in male mouse testis.Immunocytochemical and real-time PCR results showed positive Luman immunostaining in Leydig cells and the expression pattern of Luman in mouse testis exhibited an age-related increase.Immunofluorescent staining of Luman in the MLTC-1 Leydig cell line indicates that Luman is located within the cytoplasm.HCG significantly up-regulated the levels of expression of Luman in MLTC-1 cells and mouse testes,respectively.To elucidate the function of Luman, experiments were conducted to examine the consequences of lentiviral-mediated RNA interference (RNAi) of Luman on testosterone production and expression of several genes involved in steroidogenesis.Luman silencing caused a increase in HCG-stimulated testosterone production.In addition, Luman silencing significantly up-regulated the mRNA levels of P450scc and P450c17 in primary Ledyig cells and MLTC-1 cells.Overall, our data are the first to provide evidence for a possible direct action of Luman in the control of testicular function.Together, these data indicate that the expression of Luman in mouse testis is stage dependent and androgen inductive, and suggest the important role of Luman in male reproduction.

材料与方法:奶牛阴道分泌物、大肠杆菌、金黄色葡萄球菌、链球菌;观察菌落形态特征,并对菌落进行染色、菌株生理生化鉴定和16SrDNA序列分析.并将分离得到的菌株进行体外抑菌试验.结果:分离得到的Y4菌株是枯草芽孢杆菌,与GeneBank中记录的枯草芽孢杆菌同源性为99.6%.该菌株对大肠杆菌、金黄色葡萄球菌和链球菌有较好的抑菌效果.
