酱香型白酒窖池下层酒醅厌氧细菌的分离与鉴定

[目的]建立AG300厌氧工作站中厌氧细菌的分离、培养方法,并对酱香型白酒窖池下层酒醅中厌氧细菌分离、筛选、鉴定.[方法]取贵州省某白酒厂酱香型白酒下层酒醅,密封于厌氧瓶中,迅速分析试验.在厌氧工作站中对酒醅样品进行稀释涂布、平板划线,结合细菌在37℃厌氧工作站和生化培养箱中的生长情况,对厌氧细菌进行筛选.对已筛出的厌氧菌进行革兰氏染色、镜检、生理生化试验和利用16S rDNA分析方法进行分子生物学鉴定,并对其进行系统发育树的分析构建.[结论]参照《伯杰细菌鉴定手册》从酱香型白酒下层酒醅中分离出4株厌氧细菌,初步鉴定为:Clotridium xylanolyticum、Clostridium amylolyticum(丁酸梭菌)、Clostridium subterminate(近端梭菌)、Clostridiu sartagoforme(煎盘梭菌).
