  Gliomas are the most common and malignant primary brain tumors with a poor prognosis despite multiple therapies.Quercetin, a traditional Chinese medicinal herb,is an important flavonoid with the anti-cancer activity.Here we evaluated whether quercetin can inhibit glioma cell viability and migration and promote apoptosis.Treatment of three glioma cell lines with different concentrations of quercetin inhibited cell viability in a dose-dependent manner.Scratch assays indicated that quercetin significantly decreased U87-MG cell migration.Beta x-gal enzyme, DNA staining and Annexin V-EGF/PI double staining assays demonstrated promotion of cell senescence and apoptosis by quercetin.In addition, the protein levels of p-AKT,p-ERK, Bcl-2, matrix metallopeptidase 9 (MMP-9) and Fibronectin (FN) were significantly reduced when treated with quercetin.We thus conclude that quercetin may inhibit glioma cell viability, migration and promote senescence and apoptosis by suppressing Ras/MAPK/Erk and PI3K/Akt signaling pathways.Quercetin may be a potential candidate for treatment ofglioma clinically.
