靶向调控Mst1对结直肠癌SW480细胞增殖与凋亡的影响

哺乳动物不育系20样激酶1/2(mammalian sterile 20-like kinase1/2,MST1/2)是哺乳动物中Hippo信号通路的核心成员,具有调节器官大小、细胞增殖、凋亡及抑制肿瘤生长等功能.近期研究表明,Hipoo通路活性异常与结直肠癌的发生发展密切相关,在结直肠癌组织和结直肠癌来源的细胞系中YAP蛋白过表达,而通过激活MST1和MST2蛋白激酶,其可以通过抑制YAP蛋白的过表达来阻止肠道干细胞的增殖和结直肠癌的发生.然而,有关调控Mst1表达对结直肠癌细胞增殖与凋亡影响的研究尚未见报道。本研究通过分别转或靶向沉默结直肠癌SW480细胞Mst1基因,旨在观察调控Mst1表达对结直肠癌细胞增殖与凋亡的影响及其初步机制,为结直肠癌的靶向治疗提供实验依据。我们实验室采用PolyJetTM体外DNA转染试剂介导pEGFP-N1-MST1转染入结直肠癌细胞SW480,用LipofectamineTM2000转染MST1特异性-siRNA靶向沉默MST1.MTT法检测细胞存活率,流式细胞仪检测细胞凋亡,RT-qPCR和Western blot分别检测细胞MST1,PUMA,P73,P53,YAP及caspase-3 mRNA和蛋白水平。结果显示:与Control组比较,转染p-EGFP-MST1质粒组、5-FU组和p-EGFP-MST1+5-FU组细胞48h生长抑制率明显比Control组或转染p-EGFP-N1质粒组细胞高,具有显著性差异(P<0.01).此外,与转染p-EGFP-MST1质粒组或5-FU组相比较,p-EGFP-MST1+5-FU组对SW480细胞增殖抑制作用更强,统计学有显著性差异(P<0.01):p-EGFP-MST1组、p-EGFP-MST1+5-FU组细胞凋亡率均呈显著性升高(p<0.05),MST1,Phospho-YAP1(Ser127)、p73、p53、PUMA和caspase-3蛋白水平均呈显著性升高(P<0.05)、CTGF、AREG蛋白水平呈显著性降低(P<0.05);流式细胞仪检测结果显示,与Control组或NC-siRNA组细胞比较,转染MST1-siRNA3处理后,SW480细胞的总凋亡率、早期凋亡率和晚期凋亡率均降低,均具有统计学意义(P<0.01),说明沉默MST1可抑制SW480细胞凋亡。 结论:靶向调控MST1表达可较好调控结直肠癌细胞增殖与凋亡,MST1很有可能成为人结直肠癌防治的新靶点。
