目的:观察芒果苷对白血病HL-60细胞周期分布及细胞周期素B1(CyclinB1)、细胞周期分裂基因2(CDC2)表达的影响,以进一步探讨芒果苷抗白血病的分子作用机制。方法:应用MTT比色法测定芒果苷对白血病HL-60细胞增殖的抑制作用;用流式细胞术(FCM)检测芒果苷对细胞周期的影响;用RT-PCR技术检测CyclinB1及CDC2 mRNA表达水平。结果:芒果苷呈剂量及时间依赖性抑制HL-60细胞的生长,作用24 h后,G2/M期细胞增多,出现G2/M期阻滞;CyclinB1 mRNA和CDC2 mRNA的表达随药物质量浓度的增加而逐渐增强,在芒果苷质量浓度80μmol/L时表达达到峰值。结论:芒果苷可抑制HL-60细胞增殖,阻滞细胞周期于G2/M期,显著上调HL-60细胞CyclinB1 mRNA和CDC2 mRNA的表达水平。芒果苷诱导HL-60细胞G2/M期阻滞可能是其抗白血病作用的分子机制之一。
OBJECTIVE: To observe the effect of mangiferin on the cell cycle distribution, the expression of CyclinB1 and CDC2 in leukemia HL-60 cells and to explore the molecular mechanism of anti-leukemia. Methods: MTT assay was used to determine the inhibitory effect of mangiferin on the proliferation of HL-60 leukemia cells. Flow cytometry (FCM) was used to detect the effect of mangiferin on cell cycle. The expression of CyclinB1 and CDC2 mRNA was detected by RT-PCR . Results: Mangiferin inhibited the growth of HL-60 cells in a dose-and time-dependent manner. After 24 h, the number of cells in G2 / M phase increased and G2 / M phase arrest occurred. The expression of CyclinB1 mRNA and CDC2 mRNA increased with the increase of drug concentration Increased and gradually increased, the expression peaked at the concentration of mangiferin 80μmol / L. CONCLUSION: Mangiferin can inhibit the proliferation of HL-60 cells and arrest the cell cycle in G2 / M phase, and significantly up-regulate the expression of CyclinB1 mRNA and CDC2 mRNA in HL-60 cells. Mangiferin-induced G2 / M arrest in HL-60 cells may be one of the molecular mechanisms of its anti-leukemia effect.
