目的研究膀胱癌组织中抑癌基因PTEN的表达及其与微血管密度(MVD)之间的关系,评价PTEN在膀胱癌发生发展中的作用。方法应用免疫组织化学SP法检测62例膀胱癌组织和18例良性膀胱病变组织中PTEN的表达和MVD,同时结合肿瘤分期分级进行分析。结果62例膀胱癌组织中PTEN阳性率为53.23%(33/62),18例良性膀胱病变组织PTEN全部阳性表达;PTEN的表达与肿瘤分级密切相关(P<0.05);肿瘤组织MVD明显高于良性病变组织;PTEN表达与MVD呈负相关(P<0.05)。结论PTEN基因突变或缺失所导致的表达障碍可能在膀胱癌的恶性进展和血管生成过程中发挥重要的作用。
Objective To investigate the expression of tumor suppressor gene PTEN in bladder cancer and its relationship with microvessel density (MVD), and to evaluate the role of PTEN in the development of bladder cancer. Methods Immunohistochemical SP method was used to detect the expression of PTEN and MVD in 62 cases of bladder cancer and 18 cases of benign bladder lesions. At the same time, the expression of PTEN was analyzed by staging and classification. Results The positive rate of PTEN was 53.23% (33/62) in 62 cases of bladder cancer and all of 18 cases of benign bladder lesions. The expression of PTEN was closely related to tumor grade (P <0.05). The MVD in tumor tissues was significantly higher than that of PTEN The expression of PTEN was negatively correlated with MVD in benign lesions (P <0.05). Conclusions The expression of PTEN gene caused by mutations or deletions may play an important role in the malignant progression and angiogenesis of bladder cancer.
