目的研究低聚古罗糖醛酸磷酸酯对RAW264.7细胞的激活作用。方法低聚褐藻胶依次通过pH分级、高压酸解纯化的方法制备低聚古罗糖醛酸,进而采用磷酸/尿素法对其进行磷酸化修饰,制备低聚古罗糖醛酸磷酸酯。在低聚古罗糖醛酸磷酸酯对RAW264.7细胞激活实验中,分别测定了对RAW264.7细胞增殖、吞噬能力、细胞因子(TNF-α、IL-1β)释放、胞内酶(酸性磷酸酶、乳酸脱氢酶)活力的影响。结果低聚古罗糖醛酸磷酸酯能显著促进RAW264.7细胞的增殖,提高其对中性红的吞噬,促进细胞因子的释放以及增强胞内酶的活性。结论低聚古罗糖醛酸磷酸酯能够多层次、多水平的激活RAW264.7细胞,是1种潜在的免疫增强剂。
Objective To study the activation of oligoglucuronic acid phosphate on RAW264.7 cells. Methods Oligocuronic acid was prepared by the method of pH fractionation and high pressure acid hydrolysis and purification, respectively. The oligoglucuronic acid was further modified by phosphate / urea method to prepare oligoglucuronic acid phosphate ester. In the experiment of RAW264.7 cell activation by oligoglucuronic acid phosphate, the effects of proliferation, phagocytosis, release of cytokines (TNF-α, IL-1β) Phosphatase, lactate dehydrogenase) activity. Results Oligomer guluronic acid phosphate can significantly promote the proliferation of RAW264.7 cells, increase its phagocytosis of neutral red, promote the release of cytokines and enhance the activity of intracellular enzymes. CONCLUSION: Oligo guluronic acid phosphate can activate RAW264.7 cells at multiple levels and levels and is a potential immunostimulant.
