目的:探讨下丘脑-垂体-肾上腺(hypothalamic-pituitary-adrenal axis,HPA)轴系统促肾上腺皮质激素释放激素受体1(corticotropin-releasing hormone receptor 1,CRHR1)、促肾上腺皮质激素释放激素结合蛋白(corticotropin-releasing hormone binding protein,CRHBP)以及FK506结合蛋白(FK506 binding protein 5,FKBP5)基因多态性与青年人自杀未遂行为的关联性。方法:采用i MLDRTM多重SNP分型,检测71例自杀未遂青年人(自杀未遂组,年龄15~24岁)和116例健康对照(对照组)CRHR1基因(rs110402、rs7209436)、FKBP5基因(rs3800373、rs1360780)以及CRHBP基因(rs10474485、rs7728378)6个多态位点,并对基因型、等位基因及单体型频率进行比较。结果:6个位点基因型与等位基因频率在病例组和对照组中分布无统计学差异(P>0.05)。由rs110402、rs7209436位点构成的单体型,rs3800373、rs1360780位点构成的单体型,rs10474485、rs7728378构成的单体型在2组之间分布无统计学差异(P>0.05)。矫正精神疾病史后,在多个遗传模式中,rs7728378显示与自杀未遂有关联。共显性模式:C/T(OR=3.53,95%CI=1.45~8.64,P=0.011)、T/T(OR=3.40,95%CI=1.04~11.14,P=0.011);显性模式:C/T-T/T(OR=3.51,95%CI=1.46~8.43,P=0.003);超显性模式:C/T(OR=2.19,95%CI=1.08~4.45,P=0.026);累加模式:C/C、C/T及T/T(OR=1.95,95%CI=1.12~3.41,P=0.016)。结论:CRHBP基因位点rs7728378多态性与青年人的自杀未遂相关,该位点的杂合基因型C/T可能增加自杀未遂的风险。
Objective: To investigate the effects of corticotropin-releasing hormone receptor 1 (CRHR1), corticotropin-releasing hormone-binding protein The correlation between the polymorphism of FK506 binding protein 5 (FKB50) and the suicide attempt of young people was investigated. Methods: The CRHR1 gene (rs110402, rs7209436), FKBP5 gene (rs3800373, rs1360780) and CRHBP gene (rs10474485, rs7728378) polymorphic sites, and genotypes, alleles and haplotype frequencies were compared. Results: There was no significant difference in genotype and allele frequency between the 6 cases and the control group (P> 0.05). The haplotype consisting of rs110402 and rs7209436 loci, rs3800373 and rs1360780 haplotypes, rs10474485 and rs7728378 haplotypes were not significantly different between the two groups (P> 0.05). After correcting the history of mental illness, rs7728378 showed association with attempted suicide among multiple genetic patterns. The dominant modes were C / T (OR = 3.53, 95% CI = 1.45-8.64, P = 0.011) and T / T (OR = 3.40, 95% CI = 1.04-11.14, P = 0.011) (OR = 2.19, 95% CI = 1.08-4.45, P = 0.026); C / TT / T (OR = 3.51, 95% CI = 1.46-8.43, P = 0.003) Cumulative patterns: C / C, C / T and T / T (OR = 1.95, 95% CI = 1.12-3.41, P = 0.016). Conclusion: The rs7728378 polymorphism in CRHBP gene is associated with attempted suicide in young people. The heterozygous genotype C / T at this locus may increase the risk of attempted suicide.
