目的:通过检测Ack1基因在胃腺癌组织和癌旁、正常胃组织中的表达差异分析其与患者临床病例特征的相关性。方法:收集手术切除并经病理证实的胃腺癌组织石蜡标本60例作为胃癌组,癌旁组织15例及正常胃组织15例分别作为癌旁组、正常组,用免疫组织化学SP法检测胃腺癌组织与癌旁及正常组织中Ack1蛋白的表达情况,并对Ack1蛋白的表达差异及其与患者的临床病理特征用SPSS17.0软件进行统计学分析。结果:Ack1蛋白在胃癌组织中表达阳性率明显高于正常胃组织及癌旁组织。三者Ack1蛋白表达差异具有统计学意义。两两比较后认为正常胃组织与胃癌组织、胃癌组织与癌旁组织Ack1蛋白表达差异具有统计学意义,而正常胃组织与胃癌癌旁组织Ack1蛋白表达差异不具有统计学意义。再者Ack1蛋白的表达与胃癌患者的分化程度及TNM分期有关,与病人性别、年龄、肿瘤发生部位及是否淋巴结转移无关。结论:Ack1可作为一种新型胃癌标记物,在胃癌的发生及发展过程中起到重要作用,为胃癌的个体化治疗提供了实验依据。
OBJECTIVE: To analyze the association between the expression of Ack1 gene and clinical features in gastric adenocarcinoma, adjacent normal tissues and normal gastric tissues. Methods: 60 cases of gastric adenocarcinoma and paraffin-embedded specimens of gastric adenocarcinoma were collected from 60 cases of gastric adenocarcinoma, 15 cases of para-cancerous tissue and 15 cases of normal gastric tissue as adjacent normal group and normal gastric tissue respectively. Gastric adenocarcinoma was detected by immunohistochemical SP method The expression of Ack1 protein in tissues, adjacent normal tissues and normal tissues were detected. The differences of Ack1 protein expression and clinicopathological features were analyzed by SPSS17.0 software. Results: The positive expression rate of Ack1 protein in gastric cancer tissues was significantly higher than that in normal gastric tissues and paracancerous tissues. Three Ack1 protein expression differences were statistically significant. The comparison of Ack1 protein expression between normal gastric tissues and gastric cancer tissues, gastric cancer tissues and adjacent normal tissues was considered statistically significant. However, there was no significant difference in the expression of Ack1 protein between normal gastric tissues and adjacent non-cancerous tissues. Furthermore, the expression of Ack1 protein is related to the degree of differentiation and TNM staging of patients with gastric cancer, regardless of patient’s gender, age, tumor location and lymph node metastasis. Conclusion: Ack1 can be used as a new marker of gastric cancer, plays an important role in the occurrence and development of gastric cancer, and provides an experimental basis for individualized treatment of gastric cancer.
