目的:探讨α常春藤皂苷对结肠癌细胞凋亡、增殖的作用。方法:采用MTT、流式细胞仪、Western Blot检测α常春藤皂苷对结肠癌Lo Vo细胞形态、增殖、周期、凋亡的影响。结果:α常春藤皂苷改变结肠癌Lo Vo细胞形态,α常春藤皂苷4.18 mg/L组、α常春藤皂苷8.37 mg/L组、α常春藤皂苷16.73 mg/L组、奥沙利铂9.23×10-3g/L组、α常春藤皂苷8.37 mg/L+奥沙利铂9.23×10-3g/L组对Lo Vo细胞的抑制率(%)依次为:12.49±2.36,24.02±3.98,51.00±4.50,17.92±2.28,59.66±7.93组间差异显著;空白对照组、α常春藤皂苷4.18 mg/L组、α常春藤皂苷8.37 mg/L组、α常春藤皂苷16.73 mg/L组、奥沙利铂9.23×10-3g/L组、α常春藤皂苷8.37 mg/L+奥沙利铂9.23×10-3g/L组Lo Vo细胞G1期、G2期、S期百分比分别依次为:63.17±0.92,62.32±0.86,61.01±0.74,54.45±0.96,43.80±0.60,45.96±0.68;22.71±0.65,31.27±0.59,22.54±0.51,13.45±0.40,44.54±0.40,44.49±0.61;14.47±0.21,7.59±0.43,16.35±0.44,31.28±0.97,11.58±0.34,9.61±0.20,组内无显著差异。对Lo Vo细胞的凋亡率依次为:5.43±2.13,8.18±0.11,10.80±0.26,12.37±1.27,16.87±1.52,20.55±0.99,与空白对照组相比差异显著。使Lo Vo细胞各组caspase 3的相对表达量均增加,与空白对照组相比差异显著。结论:α常春藤皂苷影响结肠癌Lo Vo细胞形态、改变其周期分布,增加caspase 3表达量,促进其凋亡,抑制其增殖,与化疗药有协同作用。
Objective: To investigate the effect of α-ivy saponin on apoptosis and proliferation of colon cancer cells. Methods: MTT, flow cytometry and Western Blot were used to detect the effect of α-ivy saponin on morphology, proliferation, cycle and apoptosis of colon cancer LoVo cells. The apoptosis rate of Lo Vo cells was 5.43 ± 2.13,8.18 ± 0.11,10.80 ± 0.26,12.37 ± 1.27,16.87 ± 1.52 and 20.55 ± 0.99, which was significantly different from the blank control group. The relative expression of caspase 3 in Lo Vo cells increased, which was significantly different from that of blank control group. CONCLUSION: α ivy saponin can affect the morphology of Lo Lo cells in colon cancer, change the distribution of caspase 3, increase the expression of caspase 3, promote its apoptosis, inhibit the proliferation of colorectal cancer cells and have a synergistic effect with chemotherapy drugs.
