目的:初步观察抗共刺激分子CD 137单克隆抗体对分离培养的人外周血CD 4+CD 25+T淋巴细胞自噬、凋亡、胀亡现象的影响及其表面特征性标志物FOX p3的表达的变化。方法:采用密度梯度离心法及尼龙棉柱法分离健康志愿者外周血T淋巴细胞,磁性细胞分离器(M ACS)分离得到CD 4+CD 25+T淋巴细胞,分别利用电镜及流式细胞仪观察、检测各分组(抗CD 137单克隆抗体2μg/m l组、IgG 1同型抗体对照组)的细胞的自噬率、凋亡率、胀亡率及FOX p3的表达。结果:M ACS可成功分离CD 4+CD 25+T淋巴细胞,纯度可达80.3%~89.5%;抗CD 137单克隆抗体可促进人外周血CD 4+CD 25+T淋巴细胞自噬率及凋亡率增加;各组间的胀亡率改变及FOX p3表达差异无统计学意义。结论:抗CD 137单克隆抗体可促进CD 4+CD 25+T淋巴细胞凋亡和自噬,其作用机制未发现与胀亡显著相关,而且不影响CD 4+CD 25+T细胞中Foxp3的表达。
OBJECTIVE: To observe the effect of anti-costimulatory molecule CD 137 monoclonal antibody on the autophagy, apoptosis and expansion of CD 4 + CD 25 + T lymphocytes isolated from human peripheral blood and to detect the expression pattern of FOX p3 Changes in expression. Methods: T lymphocytes were isolated from peripheral blood of healthy volunteers by density gradient centrifugation and nylon cotton column method. CD 4 + CD 25 + T lymphocytes were isolated by magnetic cell separator (M ACS) The autophagy rate, apoptosis rate, expansion rate and FOX p3 expression in each group (2μg / ml anti-CD 137 monoclonal antibody, IgG 1 isotype antibody control group) were observed and detected. Results: The purity of CD 4 + CD 25 + T lymphocytes was 80.3% ~ 89.5% in M ACS. The anti-CD 137 monoclonal antibody could promote the autophagy rate of CD 4 + CD 25 + T lymphocytes in peripheral blood The apoptotic rate increased. There was no significant difference between the groups in the change of the rate of expansion and the FOX p3 expression. CONCLUSION: Anti-CD 137 monoclonal antibody can promote the apoptosis and autophagy of CD 4 + CD25 + T lymphocytes. The mechanism of action is not found to be significantly correlated with expansion, and it does not affect Foxp3 expression in CD 4 + CD25 + T cells expression.
