详细报道了胡萝卜抗冻蛋白基因 (afp)植物表达载体的构建过程及其直接法转化农杆菌的方法。以限制性内切酶从克隆载体上切取 afp基因 ,将其连接在相应酶切的 p BI12 1工程质粒上 ,构建成 afp植物表达载体。采用直接转化法将重组子导入根癌农杆菌 ,并用 PCR方法进行了鉴定
The process of constructing the plant expression vector of carrot antifreeze protein gene (afp) and its method of direct transformation of Agrobacterium were reported. The afp gene was excised from the cloning vector by restriction enzyme and ligated to the corresponding p BI12 1 engineered plasmid to construct the afp plant expression vector.
