目的:体外实验研究壁虎粗多肽(Gecko Crude Peptides,GCPS)诱导人肝癌细胞HepG2凋亡的机制。方法:采用MTT法检测GCPS对肝癌细胞HepG2增殖的影响;采用Hoechst33258荧光染色观察细胞凋亡的形态学改变,western-blot检测BAX和BCL-2蛋白的表达,观察壁虎粗多肽对人肝癌细胞凋亡的影响。结果:GCPS可以抑制肝癌细胞HepG2增殖,作用呈浓度和时间依赖性,其IC50为1.2 mg/mL;在GCPS的作用下,HepG2呈凋亡形态学改变,且GCPS(1.6 mg/mL、0.8 mg/mL)可以降低bcl-2的表达,升高蛋白bax的表达。结论:GCPS对HepG2细胞有增殖抑制作用,诱导其凋亡是其作用机理之一。
OBJECTIVE: To study the mechanism of apoptosis induced by Gecko Crude Peptides (GCPS) in HepG2 human hepatocellular carcinoma cells in vitro. Methods: The effect of GCPS on the proliferation of HepG2 cells was detected by MTT assay. The morphological changes of apoptosis were observed by Hoechst33258 staining. The expressions of BAX and BCL-2 protein were detected by Western-blot. The impact of death. Results: GCPS could inhibit the proliferation of HepG2 cells in a concentration- and time-dependent manner with an IC50 of 1.2 mg / mL. GCPS induced morphological changes of HepG2 cells with GCPS (1.6 mg / mL, 0.8 mg / mL) / mL) can reduce the expression of bcl-2 and increase the protein bax expression. Conclusion: GCPS can inhibit the proliferation of HepG2 cells and induce its apoptosis is one of the mechanisms.
