目的:观察分泌型 αFGF 真核表达载体转染大鼠心肌细胞后 αFGF 在体内外的分泌表达及其对宿主心肌血管形成作用的影响。方法:构建分泌型 αFGF 真核表达载体(pCDNA 3.1+SPαFGF),转染培养大鼠的心肌细胞,体外检测 αFGF 的分泌表达;将转染细胞移植入同种大鼠心肌,检测 αFGF 在体内的表达及受体心肌的血管生成。结果:硝酸纤维素膜点杂交和ELISA显示, αFGF 在转染 24-96h 细胞培养上清中有较高的表达;接受转染心肌细胞移植 96 h 后免疫组化检查发现 αFGF 在受体心肌内得到分泌表达,20 d 后组织切片发现移植心肌细胞周围的心肌中有较多的新生毛细血管。结论: 转染分泌型 αFGF 真核表达载体的心肌细胞在体内外能分泌表达 αFGF;通过心肌细胞移植,分泌的 αFGF 在受体心肌组织中有较强的血管生成作用。
OBJECTIVE: To observe the secretion and expression of αFGF in vitro and in vivo after transfection of rat cardiac myocyte with secreted αFGF expression vector and its effect on the host myocardial angiogenesis. Methods: The eukaryotic expression vector of secreting αFGF (pCDNA 3.1 + SPFGF) was constructed and transfected into cardiomyocytes of rats. The secretion of αFGF was detected in vitro. Transfected cells were transplanted into the same myocardium of rats, In Vivo Expression and Receptor Myocardial Angiogenesis. Results: Nitrocellulose membrane dot blotting and ELISA showed that αFGF was highly expressed in the supernatant of 24-96 h transfection cells. Immunohistochemical examination of transplanted cardiomyocytes revealed that αFGF was expressed in the myocardium of the recipient Secretion was expressed. Twenty days later, there were more newborn capillaries in the myocardium around the transplanted cardiomyocytes. CONCLUSION: Myocardial cells transfected with the secreted αFGF eukaryotic expression vector can express αFGF in vivo and in vitro. The secreted αFGF has a strong angiogenic effect in the myocardium of cardiomyocytes by cardiomyocyte transplantation.
