为探讨生理性深层海水(PDSW)联合热疗对人胃癌SGC-7901细胞活力和凋亡的影响,通过培养人胃癌SGC-7901细胞株,随机分为实验组、对照组和空白组,实验组加PDSW,对照组加生理盐水,空白组不做其他处理,分别在37℃、40℃和43℃下培养,其中在40℃条件下培养6 h,43℃条件下培养1 h。用MTT法检测PDSW联合热疗在不同时间点的细胞活力,同时检测细胞克隆形成率,以及运用流式细胞分析检测细胞凋亡情况。结果表明,在热疗情况下,PDSW能抑制SGC-7901细胞的增殖,影响细胞克隆的形成;同时,PDSW联合热疗(40℃6 h和43℃1 h)后能显著增加SGC-7901细胞的凋亡。以上结果表明,PDSW联合热疗可使人胃癌细胞SGC-7901活力降低,增加胃癌细胞凋亡,具有抗肿瘤作用。
To investigate the effects of physiologic deep seawater (PDSW) combined with hyperthermia on cell viability and apoptosis in human gastric cancer SGC-7901 cells, SGC-7901 human gastric cancer cell lines were randomly divided into experimental group, control group and blank group, experimental group The rats in the control group were treated with normal saline and the blank group were not treated with other treatments. The cells were cultured at 37 ℃, 40 ℃ and 43 ℃ respectively, cultured at 40 ℃ for 6 h and incubated at 43 ℃ for 1 h. The cell viability of PDSW combined with hyperthermia at different time points was detected by MTT assay. The rate of cell colony formation was also measured. The apoptosis of PDSW cells was detected by flow cytometry. The results showed that under the condition of hyperthermia, PDSW can inhibit the proliferation of SGC-7901 cells and affect the formation of cell clone. Meanwhile, PDSW combined with hyperthermia (6 h at 40 ℃ and 1 h at 43 ℃) significantly increased SGC-7901 cells Apoptosis. The above results show that, PDSW combined with hyperthermia can reduce the vitality of human gastric cancer cell SGC-7901, increase apoptosis of gastric cancer cells, with anti-tumor effect.
