目的探讨田基黄对内毒素血症引起的肝损伤的防治作用。方法对Wister大鼠用不同剂量的田基黄水煎剂或联苯双酯进行灌胃,1次/d,连续7 d。于最后一次给药1小时后,空白组尾静脉注射生理盐水,其余各组尾静脉注射LPS 5 mg/kg。在用药24小时后下腔静脉采血做实验室指标检测,取肝组织做病理学检查。结果通过肝组织切片,LPS模型组可见脂肪变性,肝细胞灶性及带状坏死,肝窭结构破坏,汇管区炎细胞浸润。而田基黄组脂肪变性少见,偶有点状坏死,无明显炎细胞浸润。表明田基黄能减轻LPS导致肝组织病理损伤,同时抑制LPS引起的小鼠血清ALT活性的比高,抑制NO,TNF-α和IL-6的过量分泌。结论田基黄时内毒素引起的肝损伤有一定的防治作用。
Objective To investigate the preventive and therapeutic effects of Tianjihuang against liver injury induced by endotoxemia. Methods Wister rats were infused with different doses of Tianhuang Huang decoction or bifendate once a day for 7 days. One hour after the last administration, saline was injected into the tail vein of the blank group, and the other groups were injected with LPS 5 mg / kg through the tail vein. 24 hours after medication, vena cava blood samples taken laboratory tests, liver tissue pathology. Results Through the liver tissue sections, the steatosis, hepatocellular foci and zonal necrosis, the destruction of the hepatic vessels and the infiltration of inflammatory cells in the portal area were observed in the LPS model group. However, Tianji yellow group had rare degeneration of the fatty tissue, occasionally necrosis and no obvious inflammatory cell infiltration. The results showed that Trithotheculum could reduce the pathological damage induced by LPS and inhibit the ALT activity in mice induced by LPS, and inhibit the excessive secretion of NO, TNF-α and IL-6. Conclusion Tiaji yellow endotoxin-induced liver injury have a preventive effect.
