目的:研究黄连解毒汤抗铜绿假单胞菌生物被膜及与阿奇霉素协同抗菌作用。方法:微量倍比稀释法测定黄连解毒汤对铜绿假单胞菌的最小抑菌浓度(MIC),棋盘稀释法测定黄连解毒汤和阿奇霉素协同抗菌作用,MTT法测定黄连解毒汤对铜绿假单胞菌生物被膜的最小抑膜浓度(SMIC),显微镜下观察药物对生物膜形态的影响。结果:黄连解毒汤对铜绿假单胞菌的MIC 100 g.L-1,阿奇霉素200 mg.L-1,两药联合后MIC分别为25 g.L-1和25 mg.L-1抑菌分级指数(FIC)0.1875,提示两药协同作用明显。黄连解毒汤对铜绿假单胞菌生物被膜的SMIC501,3 d均为15.6 g.L-1,7 d 31.25 g.L-1;SMIC801,3,7 d均为250 g.L-1,形态观察提示黄连解毒汤SMIC80浓度对铜绿假单胞菌生物被膜的抑制作用明显。结论:黄连解毒汤具有抗铜绿假单胞菌生物被膜作用,与阿奇霉素有协同抗菌作用。
Objective: To study the anti-Pseudomonas aeruginosa biofilm of Huanglian Jiedu Decoction and synergistic antibacterial effect with azithromycin. Methods: Microelement dilution ratio method was used to determine the minimum inhibitory concentration (MIC) of Huanglian Jiedu Decoction against Pseudomonas aeruginosa. The synergistic antimicrobial activity of Huanglian Jiedu Decoction and azithromycin was determined by the chessboard dilution method. The minimum inhibitory concentration (SMIC) of bacterial biofilm was observed microscopically under the influence of the drug on the biofilm morphology. Results: The inhibitory concentration of Huang Lian Jie Du Decoction against Pseudomonas aeruginosa MIC 100 gL-1, azithromycin 200 mg.L-1, the combined MIC of 25 gL-1 and 25 mg.L-1 respectively ) 0.1875, suggesting that the two drugs synergistic effect. Huanglian Jiedu Decoction on Pseudomonas aeruginosa biofilm SMIC501, 3 d were 15.6 gL-1,7 d31.25 gL-1; SMIC801,3,7 d were 250 gL-1, morphology observation prompted Huanglian Jiedu Decoction SMIC80 Concentration of Pseudomonas aeruginosa biofilm inhibition was obvious. Conclusion: Huanglian Jiedu Decoction has anti-Pseudomonas aeruginosa biofilm effect and synergistic antibacterial effect with azithromycin.
