为了寻找一种替代家兔热原试验的方法 ,最近我们对用人全血检测热原的可行性进行了初步探索。其原理是 :将细菌内毒素标准品与从健康自愿者提供的少量肝素抗凝全血共同孵育 6h后 ,用ELISA双抗夹心法测定其产生的内源性热原 (TNF -α)。试验结果表明 :TNF -α的量与内毒素的量在一定范围 (0 0 6EU mL~ 2 0 0EU mL)内有很好的相关性 ,不同来源的内毒素试验结果也是如此。说明该方法作为替代家兔热原检查的新方法有一定的可行性。
In order to find a way to replace the rabbit pyrogen test, we recently conducted a preliminary exploration of the feasibility of detecting pyrogen using human whole blood. The principle is that after incubating the bacterial endotoxin standard with a small amount of heparin anticoagulant whole blood supplied by healthy volunteers for 6 hours, the produced endogenous pyrogen (TNF-α) is measured by ELISA double-antibody sandwich method. The results showed that there was a good correlation between the amount of TNF-α and the amount of endotoxin in a certain range (0 0 6EU mL 2 0 0EU mL), as well as the endotoxin test results of different origins. It shows that this method is feasible as a new method to replace pyrogen test in rabbits.
