检测人类乳腺癌组织中Wnt信号通路中的枢纽因子-β-catenin及其下游靶分子cyclin D1的异常表达与ERβ阳性表达之间的关系。方法随机选取42例乳腺增生,58例乳腺癌病人的乳腺组织以及乳腺癌灶周围正常乳腺组织,分别应用免疫组化SP法检测β-catenin,cyclin D1的表达,应用Western-blot法测ERβ的表达。结果乳腺癌组织β-catenin呈胞质和/或胞核异位表达,其表达率为65.5%,显著高于乳腺增生和正常乳腺组织(42.9%,35.7%,P<0.05)。乳腺癌cyclinD1阳性率为74.1%,显著高于乳腺增生和正常乳腺组织(45.2%,33.3%;P均<0.01),乳腺癌ERβ阳性率为42.7%,乳腺增生组织中ERβ阳性率为80%,正常乳腺组织中ERβ阳性率为94.5%,ERβ在正常乳腺组织中的表达明显高于恶性乳腺组织,差异有统计学意义(P<0.01)。β-catenin与cyclin D1和ERβ表达分别呈明显负相关(r=-0.83,-0.87;P<0.01)。结论乳腺癌中ERβ呈低表达,而cyclinD1和β-catenin均高表达,三者呈现负相关,提示β-catenin和cyclin D1的异常表达与ERβ阳性表达之间无直接关系。
To detect the relationship between the abnormal expression of cyclin D1, a pivotal factor in the Wnt signaling pathway, and the expression of ERβ in human breast cancer. Methods 42 cases of breast hyperplasia, 58 cases of breast cancer patients and normal breast tissue around the breast lesions were randomly selected. The expression of β-catenin and cyclin D1 were detected by immunohistochemical SP method. The expression of ERβ expression. Results The expression of β-catenin in cytoplasm and / or nucleus of breast cancer tissues was significantly higher than that in normal breast tissues (42.9%, 35.7%, P <0.05). The positive rate of cyclinD1 in breast cancer was 74.1%, which was significantly higher than that in breast hyperplasia and normal breast tissue (45.2%, 33.3%, P <0.01). The positive rate of ERβ in breast cancer was 42.7% and the positive rate of ERβ in breast hyperplasia was 80% . The positive rate of ERβ in normal breast tissues was 94.5%. The expression of ERβ in normal breast tissues was significantly higher than that in malignant breast tissues (P <0.01). β-catenin was negatively correlated with the expression of cyclin D1 and ERβ (r = -0.83, -0.87; P <0.01). Conclusions ERβ is low expression in breast cancer, while cyclinD1 and β-catenin are highly expressed. There is a negative correlation between them, suggesting that there is no direct relationship between the abnormal expression of β-catenin and cyclin D1 and the positive expression of ERβ.
