科学家尝试用各种方法研制预防呼吸道合胞病毒 (RSV)感染的安全有效的疫苗。本文介绍了一种有效的预防性鼻内基因转移策略 ,即采用含编码 RSV抗原的质粒 DNA的脱乙酰壳多糖 DNA纳球〔粘膜基因表达疫苗 (MGXV)〕。在 RSV感染小鼠模型中 ,给予单剂 MGXV(2 5 μg/只 )能显著降低急性 RSV感染后的病毒滴度及病毒抗原负荷。 MGXV处理小鼠对乙酰甲基胆碱的呼吸道反应性无显著变化 ,肺部无明显炎症。这些结果证明 MGXV能预防急性 RSV感染
Scientists have tried various ways to develop safe and effective vaccines against respiratory syncytial virus (RSV) infection. This article presents an effective prophylactic strategy for intranasal gene transfer using chitosan DNA nanospheres [mucosal gene expression vaccine (MGXV)] containing the plasmid DNA encoding the RSV antigen. In a mouse model of RSV infection, administration of a single dose of MGXV (25 μg / mouse) significantly reduced virus titer and viral antigen load after acute RSV infection. There was no significant change in respiratory responsiveness to methacholine in mice treated with MGXV, and no significant inflammation in the lungs. These results demonstrate that MGXV can prevent acute RSV infection
