目的 探讨不同剂量辐射后胰岛素样生长因子Ⅱ (insulin likegrowthfactorⅡ ,IGF Ⅱ )对成骨细胞增殖的效应。方法 成骨细胞通过酶消化法从SD大鼠乳鼠颅骨中获取传代 ,分别进行 0、10 0、40 0、6 0 0、90 0cGy的γ线辐射 ,然后用含有不同浓度IGF Ⅱ的培养液培养 6d ,观察成骨细胞的增殖情况 ,并用MTT法进行检测。结果 γ线电离辐射抑制了细胞增殖 ,10 0cGy剂量时细胞的增殖率从 0辐射时的 489 1%下降到 437 5 % ,而IGF Ⅱ使增殖率恢复到完全正常的状态 ;即使到了 40 0cGy,也能使增殖率得到一定程度的恢复 ;但当剂量到达 90 0cGy时 ,IGF Ⅱ的这种促增殖效应几乎不存在。结论 IGF Ⅱ能有效拮抗电离辐射对成骨细胞的抑制效应 ,但这种作用依赖于辐射剂量的大小。
Objective To investigate the effect of insulin like growth factor Ⅱ (IGF Ⅱ) on the proliferation of osteoblasts after different doses of radiation. Methods The osteoblasts were passaged from the skull of SD rats by enzymatic digestion and irradiated with γ ray of 0, 10, 40, 60, 0 and 90 0 cGy, respectively, and then cultured in culture medium containing different concentrations of IGF Ⅱ After cultured for 6 days, the proliferation of osteoblasts was observed and detected by MTT method. Results γ-ray ionizing radiation inhibited cell proliferation. The cell proliferation rate decreased from 489 1% to 437 5% at 0 0 cGy dose, whereas IGF Ⅱ restored the proliferation rate to a completely normal state. At 40 0 cGy, But also restored the proliferation rate to a certain degree. However, when the dose reached 90 0 cGy, the proliferation effect of IGF Ⅱ was almost absent. Conclusion IGF Ⅱ can effectively antagonize the inhibitory effect of ionizing radiation on osteoblasts, but this effect depends on the size of radiation dose.
