目的:了解血清中葡萄糖-6-磷酸异构酶(glucose-6-phosphate isomerase,GPI)抗原检测在类风湿关节炎(rheumatoid arthritis,RA)患者中的诊断意义及判断其活动性的价值。方法:用酶联免疫吸附试验(ELISA)双抗体夹心法检测60例RA病人,26例其它风湿性疾病病人和100例健康人群血清中GPI抗原的浓度,所有风湿病组病例及其中50例健康人群并检测类风湿因子(RF),抗环瓜氨酸肽(CCP)抗体,RA病人同时还检测了C反应蛋白(CRP)和血沉(ESR)的浓度。结果:RA组的GPI浓度明显高于其他风湿性疾病组及健康对照组;当GPI为0.49μg/mL时,其敏感度为73.7%,特异度为92.1%;GPI与关节肿胀个数、关节疼痛个数、RF、抗CCP抗体有显著相关性,而与其他指标年龄、CRP、ESR相比均无相关性。结论:GPI在RA的诊断中具有较高特异性。GPI阳性可能与RA活动度相关。
Objective: To investigate the diagnostic value and the value of detecting the activity of glucose-6-phosphate isomerase (GPI) in patients with rheumatoid arthritis (RA). Methods: Serum levels of GPI antigen in 60 patients with RA, 26 patients with other rheumatic diseases and 100 healthy persons were detected by enzyme-linked immunosorbent assay (ELISA) double antibody sandwich method. All rheumatism patients and 50 healthy people Patients were also tested for rheumatoid factor (RF), anti-cyclic citrullinated peptide (CCP) antibodies, and RA patients were also tested for C-reactive protein (CRP) and erythrocyte sedimentation rate (ESR) concentrations. Results: The GPI level in RA group was significantly higher than that in other rheumatic diseases group and healthy control group. The sensitivity and specificity of GPI was 0.49μg / mL and 73.7% respectively, and the specificity was 92.1% There was a significant correlation between the number of pain, RF, anti-CCP antibody, but no correlation with age, CRP and ESR. Conclusion: GPI has higher specificity in the diagnosis of RA. GPI positive may be related to RA activity.
