目的:观察芹菜素对炎性因子白细胞介素6 (interleakin-6,IL-6)刺激的中性粒细胞(neutrophils,Neu)凋亡的影响,并探讨可能的分子机制.方法:采取健康志愿者外周静脉血,分离纯化Neu,随机分为对照组、芹菜素组、IL-6组、IL-6+芹菜素组及加入广谱天冬氨酸特异性半胱氨酸蛋白酶(caspase)抑制剂的Q-VD-Oph+IL-6+芹菜素组、z-VAD-fmk+IL-6+芹菜素组.流式细胞术观察Neu凋亡率,Western blot检测Neu活化的caspase-3(cleaved caspase-3)的表达情况.结果:与对照组比较,芹菜素促进Neu凋亡(P<0.05),IL-6抑制Neu凋亡(P<0.05);两者共孵育时,芹菜素可以部分逆转IL-6诱导的Neu凋亡延迟(P<0.05);与IL-6组比较,芹菜素与IL-6共孵育组cleaved caspase-3的蛋白表达水平显著增高(P<0.05),而且芹菜素对caspase-3的活化作用和对IL-6诱导的Neu凋亡延迟的逆转作用可以被caspase抑制剂消除(P<0.05).结论:芹菜素可能通过caspase途径逆转IL-6诱导的Neu凋亡延迟,为今后治疗Neu炎症性气道疾病提供了新思路.“,”Objective:To investigate the inhibitory effects of apigenin on intedeukin 6 (IL-6)-induced delay in neutrophil apoptosis and its underlying mechanisms.Methods:Neutrophils (Neu) isolated from healthy volunteers were randomly divided into the control group,apigenin group,IL-6 group,IL-6 +apigenin group,QVD +IL-6 +apigenin group,and z-VAD +IL-6 +apigenin group.Neu apoptosis was detected by flow cytometry.The protein expression of cleaved caspase-3 was determined by Western blotting analysis.Results:Compared with the control group,apigenin significantly induced Neu apoptosis (P<0.05),and IL-6 significantly inhibited Neu apoptosis (P<0.05).Apigenin partially reversed the anti-apoptotic effect of IL-6 on Neu (P<0.05).Compared with the IL-6 group,cleaved caspase3 expression was significantly increased in the IL-6+apigenin group.Moreover,caspase inhibitors QVD-Oph and z-VAD reduced the levels of cleaved caspase-3 and abolished the ability of apigenin to counteract IL-6-induced delay in Neu apoptosis.Conclusion:Apigenin overrides IL-6-induced delayed apoptosis of Neu in a caspase-dependent manner,which provides a promising target for future therapy of neutrophilic inflammatory airway disorders.
