Cyclosporine A (CyA) is an immunosuppressive agent, but nephrotoxic, and often cause high blood pressure, so often need antihypertensive treatment, its application is limited. One of the possible mechanisms of these two major adverse reactions is associated with the reactivity of CyA to enhance vascular smooth muscle stimulation of vasoconstrictors. According to Bossal-Ler and other studies, CyA induced vascular tension increase is due to endothelial dysfunction weakened endothelial protection against vasoconstrictor stimulation. Rats given high doses of CyA, its isolated aorta generated prostacyclin reduction, endothelial relaxation factor (EDRF) -mediated response was weakened. Pentoxifylline (PTX) is a drug used to treat peripheral vascular disease and to improve hemorrheology, with beneficial effects associated with increased prostacyclin synthesis and possibly enhanced EDRF-mediated responses.
