目的:探讨狗肝菜不同相对分子质量多糖对大鼠D-半乳糖胺(D-GlaN)致急性肝损伤的保护作用。方法:采用透析的方法将狗肝菜多糖分为<6 kD,>6 kD<10 kD(6 kD~10 kD),>10 kD,以及总多糖组。D-GlaN(500 mg.kg-1)ip诱导大鼠急性肝损伤模型,动物处死后测定血清中天冬氨酸转氨酶(AST),丙氨酸转氨酶(ALT)活性,制备肝组织匀浆测定其中丙二醛(MDA)、谷胱甘肽过氧化酶(GSH-Px)含量,取肝、脾脏称重,计算脏器指数,对数据进行统计分析,并观察大鼠肝脏组织形态学变化。结果:与模型组比较,狗肝菜总多糖和相对分子质量<6 kD给药组能显著降低大鼠血清中AST,ALT活性(P<0.01);各给药组均能降低肝匀浆MDA含量(P<0.01或P<0.05),均能有效升高肝组织匀浆中GSH-Px活性(P<0.01或P<0.05),降低肝脏指数(P<0.01或P<0.05),减轻肝脏病理损伤。结论:狗肝菜各多糖给药组均有一定保肝作用,且以相对分子质量<6 kD效果最佳。
AIM: To investigate the protective effect of different molecular weight polysaccharides of Dogao Gan on acute hepatic injury induced by D-galactosamine in rats. Methods: Hepatic polysaccharides were divided into 6 kD, 6 kD <10 kD (6 kD ~ 10 kD),> 10 kD, and total polysaccharide by dialysis. D-GlaN (500 mg.kg-1) ip induced acute liver injury in rats. After the animals were sacrificed, the activity of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) The contents of malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) were measured. The liver and spleen were weighed and the organ index was calculated. The data were analyzed statistically and the morphological changes of liver were observed. Results: Compared with the model group, the total hepatic polysaccharide and molecular weight of <6 kD in HepG-liver could significantly decrease the activity of AST and ALT in serum of rats (P <0.01), and the content of MDA in liver homogenate decreased P <0.01 or P <0.05), all of which can effectively increase the activity of GSH-Px in liver homogenate (P <0.01 or P <0.05), reduce the index of liver (P <0.01 or P < . Conclusion: The polysaccharides from the liver of Hepatica hepatica all have certain hepatoprotective effects, and the best results are obtained when the relative molecular mass is less than 6 kD.
