用天蚕抗菌肽B基因转化广霍香,以期产生抗广霍香青枯病的转基因株系。通过农杆菌介导的叶盘法将抗菌肽B基因转入广霍香的组织细胞,在Kan的筛选培养基上经过丛芽途径获得转基因植株;采用DNADotblot、Southernblot、RNADotblot和大田攻毒试验证实抗菌肽B基因已整合到再生植株的核基因组中,获得了高水平的表达,产生较强的抗病效果。
With silkworm Antimicrobial Peptide B gene transformation Guanghuo Xiang, in order to produce resistance to broad-leaved bacterial wilt of the transgenic lines. Agrobacterium tumefaciens-mediated leaf disc method was used to transfer the antibacterial peptide B gene into the broad-leaved tissue cells, and the transgenic plants were obtained through the bud-shoot pathway on the screening medium of Kan. Antibacterial activity was confirmed by using DNA Dotblot, Southern blot, RNA Dot blot and field- Peptide B gene has been integrated into the nuclear genome of regenerated plants to obtain a high level of expression, resulting in a stronger resistance to disease.
